The 54-year-old looks to end the club's 45-year wait for Irish Cup success in his first season in charge of the north Belfast side.
In his way is Healy, who has been at the helm at Linfield since 2015, having previously won the Irish Cup in 2017 and 2021.
The 44-year-old has also won five league titles and three League Cups at the South Belfast side, who have won the trophy a record 44 times.
He has come out on top over Magilton's side in the league meetings between the two this season, having won twice and drawing once last weekend in the previous game.
Linfield were pipped to the league title by Larne for the second consecutive season, but they did win the League Cup, beating Portadown 3-1 in the final.
Both Magilton and Healy have been nominated for the Irish Premiership Manager of the Year award and were former Northern Ireland team-mates.
Magilton made 315 appearances and scored 21 goals as a player for the Blues, before taking charge of more than 100 games as a manager at Portman Road.
Healy scored one goal in 12 games for Town on loan from Sunderland during the second of the 2009/10 season.
The game will be played at Windsor Park, with more than 15,000 supporters expected to attend the fixture, which kicks off at 2.30pm.
